how can I kill a large Euphorbia without contaminating the ground, as I want to plant something else in its place.

Cut it down first and give it a few days before digging it up. Be careful when you remove the cut stems. The milky sap that appears in cuts is highly irritant to skin (and eyes!)

Dig it all up. Euphorbias don't grow huge in my experience but they do spread so you have to make sure you've got every bit out.
